Subject: Aquatic toxicity monitoring second quarter update 2012-2013
The Aquatic Toxicology Unit (ATU) serves the state of North Carolina through its support of the EPA mandated
National Pollutant Discharge Elimination System (NPDES) as it pertains to the Clean Water Act (CWA) established.
in 1,972, ATU employs NPDES compliance and enforcement reviews, biological laboratory certification and Whole
Effluent Toxicity (WET) monitoring in its effort to support this program.
ATU has made a commitment to EPA to monitor 10% of major and 5% of minor facilities in North Carolina for
2012-2013 with an overall goal of monitoring all major facilities_ within a 10-year neiiod.. The monitoringrequire meta for each regional office appears in the table: below.

Sampling instructions
1. Please indicate split tests on the sample collection forins and remember to include the identity of helabci�nevg
the split. This is critical in order for ATU to identify and evaluate the split samples. Also ensure that the sample
collection form submitted to ATU and the, chain of custody submitted to the contract lab has matching sample type
(composite vs. grab, type of composite), sampling times, and sample collector information. ATU uses only "trite"
splits for certified laboratory evaluations. Tnie splits originate from a single grab or composite sample and are
split into 2 representative aliquots for analysis by ATU and the other- laboratory. Regional office staff should
make every effort to perform the sample split themselves or, at the very least, be present Nvben the sample is split
by facility staff unless extenuating circumstances (i.e, late sample collection times, etc.) would make it impractical.
It should be noted on the sample collection form if the regional office staff is not present at the time of the sample
split.
2. With the adoption of the new chromic; toxicity sampling schedule on January 1", 2011, samples may now be
collected earlier than 1000 hours. We do request notification, however, if a sample is to be collected earlier than
0600 hours. The maximum holding time for chronic toxicity samples is 36 hours for first use and 72 hours from
first use for test renewals The maximum holding time is 36 hours for acute toxicity samples. The holding
time for composite scruples begins at the time of completion of the 24-fir composite.
3. If a facility representative is present at the sample collection, print the name of the facility representative and
identity them as a facility representative on the sample collection form. The facility representative should also
sign the forin.
4. If the sample is a tined composite, mark the number of samples collected per hour, if the sample is a flow
proportional composite, indicate this on the sheet.
5. Its the conirnents section, indicate whether the sample was; collected using the regional office staff equipment or
the facility collection equipment.
6. sample collection material may be tempered glass, polyethylene, pertluorocarbon plastics including Tefion'�, 304
or 316 stainless steel, polypropylene, polyvinyichloride, Tygon`11; or silicone. All non-perfluorocarbon plastics
should be discarded after use. DWQ staff should not reuse non-perfiuorocarbon plastics (polypropylene, LDPE,
IIDP , polyvinyleMoride, etc.) at different facilities for collecting effluent samples. Tempered glass and
perfluorocarbon (Teflon"') plastics may be reused after bioassay cleaning.
7. All samples should be iced during collection and transport to ensure a sample holding temperature of <6°C. Be
stye to completely fill the coolers with ice and use large coolers whenever possible. This is particularly crucial for
dripping samples during warm weather months. Also, please notify, ATU staff if samples are being mailed in
multiple coolers.
